| 背景信息 | Nuclear factor of activated T cells 5 (NFAT5), also known as TonEBP, is a multidomain transcription factor that serves as the primary regulator of the cellular response to hypertonic osmotic stress. Unlike other NFAT family members, NFAT5 functions as a homodimer and is regulated by ionic strength rather than calcium signaling. It features an intrinsically disordered N-terminal domain that undergoes a conformational shift upon osmotic challenge, facilitating the activation of genes involved in osmolyte uptake, such as aldose reductase, and the expression of heat shock proteins. The C-terminal domain enables the formation of biomolecular condensates that drive nuclear transactivation. NFAT5 is essential for maintaining cellular viability in the kidney and modulating inflammatory cytokine production in immune cells. Clinically, it is associated with renal dysfunction and plays a role in autoimmune inflammation and cancer cell proliferation. |
