| 背景信息 | HEXIM P-TEFb complex subunit 1 (HEXIM1) is a transcriptional regulator that inhibits RNA polymerase II elongation by sequestering the positive transcription elongation factor b (P-TEFb). This inhibition is mediated by the binding of HEXIM1 to 7SK snRNA, which facilitates the formation of a ribonucleoprotein complex that suppresses Cdk9 kinase activity. HEXIM1 also functions as an RNA-controlled hub in the innate immune response, interacting with NEAT1 RNA in paraspeckles to form complexes with DNA-PKc and TBK1. Structurally, it contains a basic arginine-rich region for RNA binding and a C-terminal coiled-coil domain for homodimerization and Cyclin T1 interaction. Clinically, HEXIM1 acts as a tumor suppressor, and its dysregulation is associated with NUT midline carcinoma and certain immune deficiency diseases. |
