| 背景信息 | BAG cochaperone 2 (BAG2) is a cytosolic co-chaperone that functions as a nucleotide-exchange factor (NEF) for HSP70 and HSC70. It contains a C-terminal BAG domain that binds the ATPase domain of these chaperones to promote ADP release and modulate client protein folding. BAG2 also regulates protein quality control by inhibiting the E3 ubiquitin ligase CHIP, thereby suppressing the degradation of chaperone-associated clients and promoting the maturation of proteins such as CFTR. Through its involvement in the ubiquitin-proteasome system and chaperone-mediated folding, BAG2 influences the clearance of misfolded proteins. It is implicated in the mechanisms of neurodegenerative diseases and cancer, where it affects tumor cell survival and the aggregation of aberrant proteins. |
