| 背景信息 | Nudix hydrolase 3 (NUDT3) is an enzyme belonging to the Nudix superfamily that functions as a homeostatic checkpoint in nucleoside phosphate metabolism. It primarily acts as a phosphohydrolase, cleaving diphosphate groups from substrates such as diadenosine polyphosphates and 5-phosphoribose 1-diphosphate. Beyond its role in nucleotide metabolism, NUDT3 serves as an mRNA decapping enzyme that destabilizes specific transcripts, including those encoding integrin beta 6 and lipocalin-2, thereby regulating cell motility and fibronectin expression. Its activity is strictly dependent on divalent cations such as magnesium or manganese. Clinically, NUDT3 is associated with Seckel syndrome. In oncology, it is characterized as a potential tumor suppressor; for instance, its knockdown in breast cancer cells leads to the stabilization of pro-motility mRNAs and increased cellular migration. |
