| 背景信息 | Phosphoglycerate kinase 1 (PGK1) is a monomeric glycolytic enzyme of approximately 45 kDa composed of 400-415 amino acids that catalyzes the reversible conversion of 1,3-diphosphoglycerate to 3-phosphoglycerate, producing ATP in the process. The enzyme exhibits a distinctive bilobed structure with two domains of nearly equal size separated by a deep cleft and linked by flexible alpha-helices, enabling characteristic hinge-bending conformational changes. Each domain contains a Rossmann fold with a six-stranded parallel beta-sheet core; the N-terminal domain binds 1,3-bisphosphoglycerate while the C-terminal domain binds MgADP or MgATP. The catalytic mechanism involves domain closure upon substrate binding, positioning substrates for phosphoryl group transfer via nucleophilic attack. Beyond its metabolic role, PGK1 functions in cancer pathways where it promotes oncogenic signaling through AKT/mTOR activation and resists ubiquitin-mediated degradation via interactions with long non-coding RNAs, contributing to metastasis and chemoresistance in hepatocellular carcinoma and gallbladder carcinoma. |
