| 背景信息 | TIMP metallopeptidase inhibitor 1 (TIMP1) is a 28 kDa secreted glycoprotein encoded by the TIMP1 gene, belonging to the TIMP family of proteins that regulate extracellular matrix (ECM) turnover by inhibiting matrix metalloproteinases (MMPs), particularly MMP-1, -2, -3, -7, -9, and -13. TIMP1 binds reversibly to MMPs via its N-terminal domain, blocking their proteolytic activity, and also exhibits growth-promoting and anti-apoptotic properties through interactions with cell-surface receptors like CD63 and integrins. Elevated TIMP1 levels are implicated in pathological conditions, including cancer and fibrosis. In cancers (e.g., breast, colorectal), TIMP1 overexpression correlates with tumor progression, metastasis, and poor prognosis due to its dual role in ECM stabilization and promotion of cell survival. In fibrosis (e.g., liver cirrhosis, pulmonary fibrosis), excessive TIMP1 disrupts ECM homeostasis, leading to tissue scarring by reducing MMP-mediated degradation of collagen. |
