| 背景信息 | Platelet derived growth factor subunit B (PDGFB) is a secreted growth factor that plays a central role in mesenchymal cell proliferation, angiogenesis, and embryonic development. It functions as either a homodimer (PDGF-BB) or a heterodimer with PDGFA (PDGF-AB), signaling through the PDGFR-alpha and PDGFR-beta tyrosine kinase receptors. These interactions are critical for pericyte recruitment and vascular stability. Clinically, PDGFB is well-established in the pathogenesis of several disorders. Chromosomal rearrangements resulting in the COL1A1-PDGFB fusion drive constitutive autocrine signaling in dermatofibrosarcoma protuberans. Furthermore, dysregulated PDGFB signaling is a known cause of primary familial brain calcification, a condition characterized by abnormal calcium deposits in the brain vasculature and pericytes. |
