| 背景信息 | Parathyroid hormone (Parathyroid Hormone) is a critical peptide hormone secreted by the parathyroid glands to maintain systemic calcium and phosphate homeostasis. It exerts its biological effects by binding to the G-protein-coupled receptor PTH1R, which triggers the activation of adenylate cyclase and elevates intracellular cAMP levels. The hormone stimulates bone resorption via osteoclast activation and enhances renal calcium reabsorption while promoting phosphate excretion. Additionally, it indirectly increases intestinal calcium absorption by stimulating the synthesis of 1,25-dihydroxyvitamin D. Clinical dysregulation of this hormone is well-documented; primary hyperparathyroidism leads to hypercalcemia and bone loss, while hypoparathyroidism results in hypocalcemia and tetany. Defects in the PTH receptor cause pseudohypoparathyroidism, a condition characterized by impaired hormone signaling despite normal or elevated hormone levels. |
