| 背景信息 | Major histocompatibility complex, class II, DR alpha (HLA-DRA) encodes the invariant alpha chain of the HLA-DR heterodimer, a critical MHC class II molecule. Expressed on professional antigen-presenting cells such as B cells, dendritic cells, and macrophages, HLA-DRA pairs with various polymorphic beta chains to form functional receptors that present exogenous peptides to CD4+ T cells. This process is essential for the initiation and regulation of the adaptive immune response. Unlike the highly polymorphic beta chains, the HLA-DRA subunit is remarkably conserved, providing a stable framework for the peptide-binding groove. Clinically, variations in the HLA-DR complex are strongly associated with numerous autoimmune and infectious diseases, including rheumatoid arthritis and multiple sclerosis. Furthermore, high expression of HLA-DRA in the tumor microenvironment has emerged as a significant biomarker for predicting favorable responses to anti-PD-1 immunotherapy in non-small cell lung cancer. |
