| 背景信息 | Apolipoprotein E (APOE) is a multifunctional protein essential for lipid metabolism and neurobiological homeostasis. It is composed of an N-terminal domain responsible for LDL receptor binding and a C-terminal domain that initiates interactions with lipids and amyloid-beta. Synthesized primarily in the liver and brain, APOE mediates the transport and internalization of cholesterol, fat-soluble vitamins, and other lipids via cell-surface receptors. The protein exists in three major isoforms—apoE2, apoE3, and apoE4—which exhibit distinct structural topologies and functional efficiencies. Clinically, APOE is a major genetic determinant of Alzheimer's disease risk, with the apoE4 isoform strongly associated with increased disease susceptibility and accelerated neurodegeneration compared to other variants. |
