| 背景信息 | Apolipoprotein C-III (APOC3) is a protein that plays a key role in triglyceride metabolism and cardiovascular disease risk. Studies have shown that loss-of-function mutations in the APOC3 gene are associated with lower triglyceride levels and a reduced risk of ischemic cardiovascular disease and coronary heart disease. The mechanism by which APOC3 influences cardiovascular disease risk is multifaceted. APOC3 inhibits lipoprotein lipase and hepatic lipase, which are enzymes involved in the clearance of triglyceride-rich lipoproteins. This leads to increased triglyceride levels, which is a well-established risk factor for cardiovascular disease. Additionally, APOC3 has been found to directly promote atherosclerosis by localizing to atherosclerotic lesions and inducing inflammatory and calcification pathways. Elevated APOC3 levels have also been linked to impaired pancreatic beta-cell function and the development of type 2 diabetes. |
