| 背景信息 | Major histocompatibility complex, class I, A (HLA-A) is a major histocompatibility complex class I heavy chain that forms a functional heterodimer with beta-2-microglobulin. Expressed on nearly all nucleated cells, HLA-A plays a central role in the adaptive immune system by presenting endogenous peptides, derived from cytosolic proteolysis, to CD8+ cytotoxic T cells. The alpha1 and alpha2 domains of the heavy chain form a peptide-binding groove that accommodates 8-13 amino acid fragments, while the alpha3 domain facilitates CD8 binding. This mechanism is essential for immune surveillance against viruses, intracellular pathogens, and malignant cells. HLA-A is highly polymorphic, with variants dictating peptide specificity and influencing disease susceptibility. Clinical relevance is firmly established in the context of hematopoietic stem cell and organ transplantation, where HLA-A matching is critical to prevent graft rejection. |
