These proteins consist of an alpha-heavy chain non-covalently associated with beta-2-microglobulin. The alpha1 and alpha2 domains form a closed-ended peptide-binding groove that presents endogenous antigens to CD8+ T cells. While HLA-A, B, and C are ubiquitously expressed and highly polymorphic to ensure broad immune coverage, HLA-G is non-polymorphic and primarily restricted to trophoblasts, where it modulates immune tolerance via inhibitory receptors like LILRB1. These molecules are essential for viral defense and self-recognition. Clinical relevance is well-documented, with specific alleles linked to autoimmune conditions, such as HLA-B27 in ankylosing spondylitis, and HLA-G dysregulation associated with pregnancy complications and tumor immune evasion. |
