| 背景信息 | Thymidylate synthetase (TYMS) is a cytosolic, homodimeric enzyme that catalyzes the reductive methylation of dUMP to dTMP, utilizing 5,10-methylenetetrahydrofolate as a cofactor. This reaction provides the sole de novo source of dTMP, making TYMS essential for DNA synthesis and repair. By maintaining the thymidylate pool, the enzyme ensures balanced deoxynucleotide supply and normal cell-cycle progression. TYMS is a primary target for several antimetabolite chemotherapeutic agents, including 5-fluorouracil and folate analogs, which induce thymineless death in rapidly dividing cells. Clinically, TYMS expression levels and genetic polymorphisms are established markers for predicting response and resistance to fluoropyrimidine-based therapies. Additionally, mutations in the TYMS gene have been linked to digenic forms of dyskeratosis congenita and increased susceptibility to certain neoplastic diseases. |
