| 背景信息 | Ribophorin I (RPN1) is a type I integral membrane glycoprotein of the rough endoplasmic reticulum and a non-catalytic subunit of the oligosaccharyltransferase (OST) complex. It plays a critical role in the secretory pathway by mediating N-linked glycosylation of nascent polypeptides at Asn-X-Ser/Thr motifs. RPN1 facilitates the selection and delivery of precursor proteins to the OST core, thereby influencing glycoprotein folding, stability, and quality control. Experimental evidence indicates that RPN1 knockdown induces endoplasmic reticulum stress and triggers apoptosis. Clinically, dysregulated RPN1 expression is associated with breast cancer, where elevated protein levels correlate with adverse clinicopathologic features, poor relapse-free survival, and increased tumor cell proliferation, migration, and invasion. |
