| 背景信息 | S100 calcium binding protein A8 (S100A8), also known as calgranulin A, is a calcium- and zinc-binding protein that frequently functions as a heterodimer with S100A9 to form calprotectin. Primarily expressed in neutrophils and monocytes, it regulates intracellular processes such as the cell cycle, cytoskeleton dynamics, and NADPH-oxidase activation. When secreted into the extracellular space, S100A8 acts as a damage-associated molecular pattern (DAMP) or alarmin, binding to TLR4 and AGER (RAGE) to trigger NF-κB and MAPK signaling pathways. This activity amplifies pro-inflammatory cascades and facilitates antimicrobial activity through metal ion chelation. S100A8 is a well-established biomarker and mediator in inflammatory diseases, including rheumatoid arthritis, inflammatory bowel disease, and cystic fibrosis. It also plays a significant role in the tumor microenvironment, where its expression correlates with poor prognosis in various cancers. |
