| 背景信息 | Protein kinase C beta (PKC-beta 1) is a conventional serine/threonine kinase that mediates a wide array of cellular signaling processes, including B-cell activation, endothelial cell proliferation, and glucose metabolism. It is activated by the secondary messengers diacylglycerol (DAG) and calcium, which trigger its recruitment to the plasma membrane. The enzyme's activity is further regulated by constitutive phosphorylation by PDK1 and mTORC2. PKC-beta 1 plays a critical role in the NF-kappa-B signaling pathway, particularly during the immune response, and regulates intestinal sugar absorption. Clinically, dysregulation of PKC-beta 1 is strongly associated with the development of diabetic complications, such as retinopathy and nephropathy, as well as cardiovascular disorders including heart failure. It is also implicated in the pathogenesis of B-cell lymphomas and retinoblastoma. |
