| 背景信息 | Decorin (DCN) is a small leucine-rich proteoglycan (SLRP) that plays a fundamental role in organizing the extracellular matrix. It is characterized by a leucine-rich protein core and a single glycosaminoglycan chain, which allow it to bind type I collagen and regulate fibrillogenesis. Decorin acts as a potent biological modulator by sequestering growth factors such as TGF-β, thereby inhibiting fibrotic signaling, and by interacting with receptor tyrosine kinases like EGFR and Met to suppress tumor growth and angiogenesis. It also functions as a myokine that regulates muscle hypertrophy by inhibiting myostatin. Clinically, mutations in the decorin gene cause congenital stromal corneal dystrophy, while its deficiency is associated with skin fragility. In oncology, decorin is recognized for its ability to curb cell proliferation and overcome apoptosis resistance. |
