| 背景信息 | Matrix metallopeptidase 3 (MMP3), also known as stromelysin-1, is a zinc-dependent endopeptidase involved in the degradation and remodeling of the extracellular matrix (ECM). It possesses broad substrate specificity, targeting components such as fibronectin, laminin, various collagen types (I, III, IV, V, IX, and X), and cartilage proteoglycans. Beyond direct ECM degradation, MMP3 plays a pivotal role in the activation of other pro-matrix metalloproteinases, thereby initiating a proteolytic cascade. The enzyme's activity is regulated by its propeptide domain and depends on the coordination of zinc ions within its catalytic site. MMP3 is essential for physiological processes such as wound repair and tissue remodeling. Pathologically, its dysregulation is well-established in the progression of atherosclerosis, rheumatoid arthritis, and tumor invasion, where excessive ECM breakdown facilitates disease advancement. |
