| 背景信息 | Glutathione S-transferase alpha 1 (GSTA1) is a cytosolic phase II detoxification enzyme that plays a central role in the metabolism of xenobiotics and endogenous electrophilic compounds. It catalyzes the nucleophilic attack of the glutathione sulfur atom on various substrates, facilitating their neutralization and excretion. Structurally, GSTA1 is composed of an N-terminal thioredoxin-like domain containing a conserved glutathione-binding site (G-site) and a C-terminal all-helical domain that forms a hydrophobic substrate-binding site (H-site). Beyond its primary role in detoxification, GSTA1 is involved in cellular signaling and the regulation of biosynthetic pathways. Clinically, the absence or functional impairment of GSTA1 is associated with an increased risk of several human diseases, including Parkinson’s disease, Alzheimer’s disease, and cardiovascular disorders, due to reduced capacity to manage oxidative stress and toxic metabolites. |
