| 背景信息 | MET proto-oncogene, receptor tyrosine kinase (MET) encodes a transmembrane receptor tyrosine kinase that is essential for embryonic development, wound healing, and angiogenesis. Upon binding its ligand, hepatocyte growth factor (HGF), the MET receptor undergoes autophosphorylation and recruits various adapter proteins, including GRB2 and GAB1. This activation triggers multiple downstream signaling pathways, such as MAPK, PI3K/AKT, and STAT3, which drive cellular proliferation, motility, and invasive growth. MET is a well-characterized oncogene that is frequently dysregulated in human cancers through gene amplification, overexpression, or specific mutations, such as exon 14 skipping. These alterations promote tumor progression, metastasis, and resistance to conventional therapies. Consequently, MET has become a major therapeutic target in oncology, particularly in non-small cell lung cancer and gastric carcinoma, where small-molecule inhibitors and monoclonal antibodies are utilized to disrupt its signaling. |
