| 背景信息 | POU class 2 homeobox 2 (POU2F2) is a transcription factor that specifically binds to the octamer motif (5'-ATTTGCAT-3'), a sequence commonly found in immunoglobulin gene promoters, and plays a crucial role in regulating gene expression in B cells, particularly influencing their proliferation and differentiation. As a member of the POU transcription factor family, POU2F2 is essential for immune response modulation and can regulate the expression of genes such as IL6 in B cells. Structurally, it contains a POU-specific domain and a homeobox domain, enabling precise DNA binding and transcriptional activation. Beyond its physiological roles, POU2F2 has been implicated in cancer biology, where its overexpression is associated with tumor-promoting activities, such as enhancing cell proliferation, motility, and glycolytic reprogramming, notably in glioblastoma and lung cancer. Mechanistically, POU2F2 can activate the PI3K/AKT/mTOR pathway, contributing to tumor progression and poor prognosis in certain cancers. |
