| 背景信息 | Fructose-bisphosphatase 1 (FBP1) is a rate-limiting gluconeogenic enzyme that catalyzes the hydrolysis of fructose 1,6-bisphosphate to fructose 6-phosphate and inorganic phosphate. This reaction opposes the glycolytic activity of phosphofructokinase and is essential for maintaining blood glucose levels during fasting. FBP1 functions as a tetramer and is allosterically inhibited by AMP and fructose 2,6-bisphosphate. Beyond its metabolic role in the cytoplasm, FBP1 can translocate to the nucleus to regulate gene expression and suppress non-canonical signaling pathways. Genetic deficiency of FBP1 leads to fructose-1,6-bisphosphatase deficiency, a disorder characterized by hypoglycemia and metabolic acidosis. In clinical oncology, FBP1 is frequently downregulated in various cancers, such as hepatocellular carcinoma and renal cell carcinoma, where its loss promotes the Warburg effect and tumor progression. |
