| 背景信息 | Clathrin light chain B (CLTB) is a regulatory subunit of clathrin triskelions, the primary structural components of the polyhedral lattice that coats membranes during receptor-mediated endocytosis. CLTB associates with the clathrin heavy chain (CHC17) to provide tensile strength to the lattice and prevent premature polymerization, thereby regulating the assembly and disassembly of coated vesicles. It facilitates cargo selection and vesicle formation through interactions with actin-organizing proteins such as HIP1 and HIP1R, as well as the kinase LRRK2. The protein features unique phosphorylation sites that modulate the internalization of G-protein-coupled receptors and influence lattice curvature. While CLTB is essential for maintaining efficient clathrin recycling and cellular signaling, there are currently no robustly established human diseases directly linked to mutations in the CLTB gene. Its function is primarily understood through its role in fundamental membrane trafficking. |
