| 背景信息 | GLI family zinc finger 2 (GLI2) is a transcription factor belonging to the GLI family of zinc finger proteins. It functions as a mediator of Sonic hedgehog (Shh) signaling and plays crucial roles in embryonic development and cell fate determination. GLI2 contains DNA-binding zinc finger motifs, a repressor domain at the N-terminus, and an activator domain at the C-terminus. It can act as both a transcriptional activator and repressor, with its activity regulated through post-translational modifications and protein-protein interactions. GLI2 localizes to the cytoplasm and nucleus, where it regulates target gene expression involved in proliferation, survival, and differentiation. Mutations in GLI2 are associated with holoprosencephaly, a disorder of forebrain development, as well as pituitary anomalies and polydactyly. GLI2 dysregulation has also been implicated in various cancers, including medulloblastoma and basal cell carcinoma. |
