| 背景信息 | KIT proto-oncogene, receptor tyrosine kinase (KIT) is a transmembrane protein that plays essential roles in cell survival, proliferation, and differentiation. It consists of an extracellular domain with five immunoglobulin-like domains, a transmembrane segment, and an intracellular tyrosine kinase domain. Upon binding to its ligand, stem cell factor (SCF), KIT dimerizes and activates multiple signaling pathways, including PI3K/AKT, MAPK, and JAK/STAT. KIT is crucial for the development and function of hematopoietic stem cells, melanocytes, germ cells, and interstitial cells of Cajal. Mutations or dysregulation of KIT are associated with various cancers, including gastrointestinal stromal tumors (GISTs), acute myeloid leukemia, and melanoma. In GISTs, activating mutations in KIT lead to constitutive activation and uncontrolled cell proliferation. |
