| 背景信息 | DNA topoisomerase I (TOP1) is a nuclear enzyme essential for relieving the torsional strain generated during DNA replication, transcription, and recombination. It operates by transiently cleaving one strand of the DNA duplex, forming a covalent phosphodiester bond between a catalytic tyrosine residue and the 3'-phosphate of the broken strand. This allows the intact strand to pass through the break, releasing superhelical tension before the enzyme religates the DNA. TOP1 functions independently of ATP and is characterized by a C-shaped architecture that clamps around the DNA. Clinically, TOP1 is a major target for chemotherapeutic inhibitors, which stabilize the covalent cleavage complexes to induce lethal DNA damage in rapidly dividing cancer cells. Dysregulation or mutation of TOP1 is associated with genomic instability and various malignancies. |
