| 背景信息 | Homeobox D8 (HOXD8) is a sequence-specific transcription factor belonging to the highly conserved HOXD gene cluster. It plays a fundamental role in regulating anterior-posterior positional identity and limb morphogenesis during embryonic development. The protein contains a characteristic homeodomain that enables DNA-binding activity, allowing it to regulate the expression of target genes involved in skeletal and urogenital development. While HOXD8 is expressed in various tissues during embryogenesis, its specific adult functions are still being explored. Clinically, deletions within the HOXD cluster that encompass HOXD8 are associated with severe limb malformations, including synpolydactyly and genital abnormalities. It has also been implicated in the pathogenesis of clubfoot. Although direct causative mutations isolated to HOXD8 alone are rare, its role within the HOXD cluster is critical for the proper patterning of the extremities and the development of the urogenital system. |
