| 背景信息 | Keratin 10 (Cytokeratin-10) is a type I acidic intermediate filament protein expressed in the suprabasal layers of stratified keratinizing epithelia. It forms obligatory heterodimers with type II keratin K1, assembling into higher-order filaments that provide mechanical strength and toughness to the epidermis. The protein features a conserved tri-domain architecture, including a central alpha-helical rod domain stabilized by hydrophobic interactions and disulfide cross-links, which significantly enhance the structural integrity of the skin. These filaments are essential for epithelial differentiation and barrier function. Clinically, missense mutations in the rod domain of KRT10 disrupt filament assembly and dimer stability, leading to epidermolytic ichthyosis. This severe genodermatosis is characterized by skin fragility, blistering, and hyperkeratosis resulting from cytolysis in the basal and suprabasal layers of the epidermis. |
