| 背景信息 | CEA cell adhesion molecule 1 (CEACAM1) is a single-pass transmembrane glycoprotein expressed on epithelial cells, endothelial cells, and leukocytes. It functions as a versatile cell adhesion molecule, mediating both homophilic and heterophilic interactions to regulate cell proliferation, differentiation, and apoptosis. In the immune system, CEACAM1 acts as a key negative regulator; it forms heterodimers with molecules such as TIM-3 to promote T-cell inhibition and is a marker of T-cell exhaustion during chronic infection and malignancy. It also modulates innate immune responses and angiogenesis. Clinically, CEACAM1 is associated with autoimmunity and cancer progression, where it promotes epithelial-to-mesenchymal transition and tumor growth. Its role in immune evasion makes it a significant target in cancer immunotherapy, as co-blockade of CEACAM1 and TIM-3 has been shown to enhance anti-tumor immune responses. |
