| 背景信息 | Coagulation factor III, tissue factor (Tissue Factor) is a transmembrane glycoprotein that serves as the primary initiator of the extrinsic coagulation cascade. Upon vascular injury, it is exposed to the blood and binds with high affinity to circulating Factor VIIa. This complex subsequently activates Factor X, leading to thrombin generation and the formation of a fibrin clot. Beyond its role in hemostasis, Tissue Factor functions as a signaling receptor through its cytosolic domain, influencing processes such as angiogenesis, inflammation, and cell migration. It is expressed on subendothelial cells, fibroblasts, and activated leukocytes. Soluble isoforms generated by alternative splicing also contribute to non-hemostatic functions by interacting with integrins. Clinically, the dysregulation of Tissue Factor is strongly linked to cancer progression, particularly in pancreatic, breast, and lung carcinomas. |
