| 背景信息 | Major histocompatibility complex, class I, E (HLA-E) is a non-classical MHC class I molecule characterized by limited polymorphism and low cell surface expression. It exists as a heterodimer composed of a heavy chain and beta-2 microglobulin. HLA-E primarily binds a restricted subset of conserved leader sequence peptides derived from classical MHC class Ia molecules. These peptide-MHC complexes are presented to NK cell receptors, specifically the CD94/NKG2 family. Interaction with CD94/NKG2A or CD94/NKG2B delivers inhibitory signals that prevent NK cell-mediated cytotoxicity, allowing the immune system to monitor healthy MHC class I expression. Conversely, engagement with CD94/NKG2C can trigger NK cell activation. Beyond its role in NK cell regulation, HLA-E can present pathogen-derived peptides to CD8+ T cells, facilitating targeted antimicrobial immune responses. |
