| 背景信息 | CD99 molecule (Xg blood group) (CD99) is a heavily O-glycosylated type I transmembrane glycoprotein expressed on leukocytes, thymocytes, and endothelial cells. It exists in long and short isoforms generated through alternative splicing. CD99 plays a critical role in leukocyte transendothelial migration, where it works alongside PECAM-1 to facilitate the final step of extravasation across the endothelial barrier. It also mediates T-cell adhesion and triggers T-cell death through caspase-independent pathways. Recent structural evidence highlights unique dimerization interfaces that support its homotypic binding properties. Clinically, CD99 is highly relevant in hematological malignancies and has been identified as a synthetic lethal partner with MYC in specific lymphomas. It also serves as a diagnostic marker and functional player in other malignancies, including osteosarcoma and Hodgkin's lymphoma. |
