| 背景信息 | Interleukin 1 receptor type 1 (IL1R1) is a high-affinity cell surface receptor essential for mediating the biological effects of the proinflammatory cytokines IL-1 alpha and IL-1 beta, as well as their antagonist, IL-1Ra. The receptor's extracellular region contains three immunoglobulin-like domains that form a binding pocket for IL-1 ligands. Upon ligand binding, IL1R1 recruits the accessory protein IL-1RAcP to form a heterotrimeric signaling complex. This assembly triggers the activation of intracellular pathways, including NF-kappaB, MAPK, and MyD88-dependent signaling, which are critical for innate and adaptive immunity and Th1 interferon-gamma costimulation. Clinically, dysregulated IL1R1 signaling is a central driver of various autoinflammatory conditions and chronic inflammatory diseases, most notably rheumatoid arthritis, where it promotes persistent tissue inflammation and damage. |
