| 背景信息 | Vascular endothelial growth factor A (VEGFA) is a glycosylated, disulfide-linked homodimeric protein belonging to the PDGF/VEGF growth factor family. It acts as a potent mitogen primarily on endothelial cells to induce proliferation, promote migration, inhibit apoptosis, and increase vascular permeability. VEGFA exerts its biological effects by binding to the tyrosine kinase receptors VEGFR1 and VEGFR2, triggering signaling cascades such as the phospholipase C-gamma and MAPK pathways. Beyond its primary roles in angiogenesis and vasculogenesis, VEGFA serves as a chemoattractant for osteoblasts during bone remodeling and facilitates wound repair through the formation of granulation tissue. Pathologically, VEGFA is essential for sustaining tumor angiogenesis and cancer stem cell functions. Clinically, dysregulation of VEGFA is a critical driver of glioblastoma progression and various vascular disorders, making it a major therapeutic target in oncology and ophthalmology. |
