| 背景信息 | CTLA-4 (Cytotoxic T-Lymphocyte Antigen-4) is a crucial negative regulator of T-cell activation and plays a pivotal role in maintaining immune homeostasis and self-tolerance. Dysregulation or deficiency of CTLA-4 leads to uncontrolled T-cell activation and proliferation, resulting in a breakdown of self-tolerance and susceptibility to various autoimmune diseases such as rheumatoid arthritis, systemic lupus erythematosus, multiple sclerosis, type 1 diabetes, and myasthenia gravis. Conversely, polymorphisms or mutations in the CTLA-4 gene have been associated with an increased risk of autoimmune disorders. CTLA-4 plays a crucial role in regulating anti-tumor immunity, and its blockade through checkpoint inhibitors like ipilimumab has shown promising results in cancer immunotherapy. |
