| 背景信息 | Thyroid stimulating hormone receptor (TSHR) is a G-protein-coupled receptor primarily located on the plasma membrane of thyroid follicular cells. It consists of a large extracellular domain with leucine-rich repeats, seven transmembrane helices, and an intracellular C-terminal tail. Upon binding thyroid-stimulating hormone (TSH), the receptor undergoes conformational changes that activate Gs and Gq protein subtypes, triggering cAMP and phospholipase C signaling cascades. These pathways regulate critical thyroid functions, including iodide uptake, thyroid peroxidase expression, and the synthesis of thyroglobulin. TSHR is central to the pathogenesis of several thyroid disorders. In Graves' disease, autoantibodies act as agonists to constitutively stimulate the receptor, leading to hyperthyroidism. Conversely, blocking antibodies are associated with Hashimoto's thyroiditis. Additionally, mutations that cause constitutive activation of TSHR are linked to toxic thyroid adenomas, and its expression levels are often monitored in the context of thyroid carcinoma management. |
