| 背景信息 | X-box binding protein 1 (XBP1) is a basic leucine zipper transcription factor that serves as a master regulator of the unfolded protein response (UPR) and endoplasmic reticulum (ER) stress. Upon ER stress, XBP1 mRNA undergoes unconventional splicing by IRE1 to produce the active XBP1s isoform, which upregulates ER chaperones, components of ER-associated degradation, and phospholipid biosynthesis to restore proteostasis. Beyond its role in the UPR, XBP1 is essential for plasma cell differentiation, immunoglobulin production, and the development of secretory tissues. Clinically, dysregulated XBP1 signaling is a hallmark of multiple myeloma survival and is implicated in atherosclerosis, neurodegenerative diseases, and metabolic disorders. In cancer, persistent XBP1 activation promotes tumor growth, angiogenesis, and resistance to therapy. |
