| 背景信息 | Fms related receptor tyrosine kinase 1 (FLT1), also known as VEGFR1, is a cell-surface tyrosine kinase receptor that binds VEGFA, VEGFB, and placental growth factor (PlGF). It is essential for embryonic vasculature development and is expressed primarily on endothelial cells and macrophages. While FLT1 possesses a much higher affinity for VEGF than VEGFR2, its intrinsic kinase activity is significantly lower, allowing it to function primarily as a VEGF trap that regulates ligand availability and prevents excessive endothelial proliferation. It modulates cell migration and survival through interactions with partners like PTK7 and the activation of Akt and FAK pathways. Clinically, dysregulation of FLT1 is implicated in pathological angiogenesis in cancer and various vascular disorders. Additionally, certain genetic variants of FLT1 are associated with reduced hypertension-related mortality and increased longevity. |
