| 背景信息 | TNF receptor superfamily member 1A (TNFRSF1A) encodes tumor necrosis factor receptor 1 (TNFR1), a ubiquitous transmembrane receptor that mediates the effects of TNF-alpha and lymphotoxin-alpha. Upon ligand binding, the receptor trimerizes and recruits intracellular adapter proteins such as TRADD, TRAF2, and FADD. This assembly triggers two primary signaling pathways: the activation of NF-kappaB, which promotes inflammation and cell survival, or the induction of caspase-mediated apoptosis through the death-inducing signaling complex. A soluble form of the receptor, produced via proteolytic shedding, acts as a natural antagonist to modulate TNF activity. Clinically, mutations in TNFRSF1A are the established cause of tumor necrosis factor receptor-associated periodic syndrome (TRAPS), an autosomal dominant autoinflammatory disorder characterized by recurrent fevers, abdominal pain, and skin rashes. Additionally, variations in this receptor are linked to an increased susceptibility to chronic inflammatory conditions like multiple sclerosis. |
