| 背景信息 | Transcription factor binding to IGHM enhancer 3 (Transcription Factor E3), or TFE3, is a basic helix-loop-helix transcription factor that serves as a master regulator of lysosomal biogenesis and the immune response. It functions by binding to E-box and CLEAR-box sequences within gene regulatory regions. TFE3 activity is tightly controlled by mTOR signaling, where nutrient-rich conditions promote its phosphorylation and cytoplasmic sequestration, while starvation triggers its dephosphorylation and nuclear translocation. It plays vital roles in maintaining stem cell pluripotency, regulating adipose tissue browning, and supporting T-cell-dependent immunity. Clinically, TFE3 is associated with X-linked intellectual developmental disorders. Furthermore, chromosomal translocations involving the TFE3 gene and partners such as PRCC or ASPSCR1 are well-established drivers of renal cell carcinoma and other malignancies, where the resulting fusion proteins promote oncogenic transformation. |
