背景信息Thymidine phosphorylase (TYMP), also known as platelet-derived endothelial cell growth factor, is a crucial enzyme involved in pyrimidine metabolism, specifically catalyzing the reversible phosphorolysis of thymidine to thymine and 2-deoxyribose-1-phosphate. The enzyme's properties include its dimeric structure and specific binding sites that facilitate its catalytic function. This protein, which functions as a homodimer, plays a vital role in maintaining nucleoside homeostasis necessary for DNA repair and replication, with its activity being essential for mitochondrial DNA integrity. Dysregulation or mutations in the TYMP gene are linked to several diseases, notably mitochondrial neurogastrointestinal encephalopathy (MNGIE) syndrome, characterized by mitochondrial dysfunction and severe neurological symptoms due to impaired thymidine salvage pathways.
