| 背景信息 | Mannose-6-phosphate receptor, cation dependent (M6PR) is a type I transmembrane glycoprotein that plays a central role in the trafficking of lysosomal enzymes. Functioning as a homodimer, it recognizes and binds mannose-6-phosphate (M6P) tags on acid hydrolase precursors within the trans-Golgi network. This binding is highly dependent on divalent cations, such as manganese, and occurs optimally at a slightly acidic pH. The receptor-cargo complex is transported via clathrin-coated vesicles to endosomes, where the lower pH environment triggers the release of the enzymes for subsequent delivery to lysosomes. Following cargo dissociation, M6PR recycles back to the Golgi apparatus via GGA adaptors. While it is a key component of the lysosomal targeting pathway, its specific biological role is distinct from the cation-independent receptor, and it serves as a critical model for understanding pH-dependent protein-carbohydrate interactions. |
