| 背景信息 | Insulin like growth factor binding protein 4 (IGFBP4) is the smallest member of the IGFBP family and acts as a key regulator of insulin-like growth factor (IGF) signaling. It binds both IGF-I and IGF-II with high affinity, extending their half-life in circulation while simultaneously inhibiting their interaction with the type 1 IGF receptor (IGF1R). This inhibition is achieved through a high-affinity N-terminal binding site and a C-terminal domain that provides steric hindrance, effectively blocking IGF access to its receptor. Unlike other family members, IGFBP4 is consistently identified as an inhibitor of cell proliferation. Clinically, it has been shown to suppress the growth of various malignancies, including prostate and colon cancers, by acting as an apoptotic factor and modulating the availability of IGFs within the tumor microenvironment. |
