| 背景信息 | Nuclear receptor subfamily 4 group A member 1 (NR4A1) is an orphan nuclear receptor that functions as a transcription factor. It binds to the NGFI-B response element (NBRE) and regulates gene expression involved in cell proliferation, apoptosis, and inflammation. NR4A1 can be induced by various stimuli, including growth factors and stress, and plays roles in energy homeostasis, immune regulation, and vascular response to injury. The protein can translocate from the nucleus to mitochondria, triggering apoptosis. In macrophages, NR4A1 regulates metabolic adaptations during inflammation by repressing genes involved in the citric acid cycle. NR4A1 is associated with diseases such as pseudohypoaldosteronism and pyomyositis. In cancer, NR4A1 has been implicated in colorectal cancer progression, where its depletion inhibits cancer advancement by promoting necroptosis via the RIG-I-like receptor pathway. |
