| 背景信息 | Protein tyrosine phosphatase receptor type Z1 (PTPRZ1) is a transmembrane enzyme primarily expressed in the central nervous system, where it regulates oligodendrocyte precursor proliferation, survival, and remyelination. Structurally, it contains an N-terminal carbonic anhydrase-like domain, fibronectin type III domains, and intracellular protein tyrosine phosphatase domains. PTPRZ1 functions by binding ligands such as pleiotrophin, midkine, and IL-34 via its extracellular chondroitin sulfate glycans. This interaction inhibits its phosphatase activity, leading to the stabilization and nuclear translocation of beta-catenin, which subsequently activates the transcription of genes like MYC and Cyclin D1. These pathways enhance cell migration, invasion, and epithelial-mesenchymal transition. Clinically, PTPRZ1 is overexpressed in various malignancies, including gliomas and breast, prostate, and colorectal cancers, where it drives tumor growth and is associated with poor prognosis. It also plays a significant role in demyelinating diseases such as multiple sclerosis. |
