| 背景信息 | SP100 nuclear antigen (SP100) is a major structural component of promyelocytic leukemia (PML) nuclear bodies, which are subnuclear organelles involved in gene regulation, antiviral defense, and the DNA damage response. It functions as a transcriptional corepressor and plays a significant role in restricting the replication of DNA viruses by binding to unmethylated CpG motifs in viral genomes via its SAND domain. SP100 also interacts with heterochromatin protein 1 (HP1) and the MRN complex to modulate chromatin structure and telomere stability. Its expression is strongly induced by interferons, leading to an increase in the size and number of nuclear bodies during stress or infection. Clinically, SP100 is a well-characterized autoantigen in primary biliary cirrhosis, where patients develop specific autoantibodies against the protein. Additionally, it acts as a tumor suppressor by inhibiting endothelial invasion and angiogenesis. |
