| 背景信息 | CD27 is a glycosylated type I transmembrane protein of approximately 55 kDa, belonging to the TNF-receptor superfamily, expressed on naïve and activated T cells, NK cells, B cells, and early hematopoietic stem cells, forming homodimers via a disulfide bridge in its extracellular cysteine-rich domains. It binds ligand CD70 to transduce costimulatory signals via TRAF2, TRAF3, TRAF5 adaptors, activating NF-κB and JNK pathways to promote T-cell immunity generation, memory, expansion, B-cell activation, immunoglobulin synthesis, and cytokine production like IFNγ, while also associating with proapoptotic SIVA for regulated cell death. A soluble form sheds post-T-cell activation. Diseases linked to CD27 include Lymphoproliferative Syndrome 2 and Combined T/B Cell Immunodeficiency, often from mutations like C53Y, C96Y, R107C reducing expression. |
