| 背景信息 | Tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activation protein theta (YWHAQ) encodes the 14-3-3 protein theta, a highly conserved molecular adapter. It belongs to the 14-3-3 family of proteins, which regulate diverse cellular processes by binding to phosphoserine or phosphothreonine motifs on a wide array of partner proteins. YWHAQ modulates signaling pathways involved in apoptosis, cell cycle control, protein trafficking, and metabolism. It interacts with and regulates the activity of various enzymes, including tyrosine and tryptophan hydroxylases, as well as kinases like PDPK1. Clinically, YWHAQ has been implicated in several neurological and oncogenic conditions. It is notably upregulated in the spinal cord of patients with sporadic amyotrophic lateral sclerosis (ALS) and has been linked to adult ADHD through epistatic interactions with other 14-3-3 isoforms. Additionally, its role in stabilizing oncogenic transcription factors like ETV1 suggests a contribution to the progression of certain cancers. |
