| 背景信息 | 5-hydroxytryptamine receptor 2C (HTR2C) is a class A G protein-coupled receptor for serotonin primarily expressed in the central nervous system, including the hypothalamus. It features a seven-transmembrane structure and couples to Gq/11 proteins to activate phospholipase C-beta, leading to intracellular calcium mobilization and protein kinase C activation. The HTR2C mRNA undergoes extensive RNA editing and alternative splicing to produce isoforms with varying constitutive activities. Functionally, the receptor modulates dopaminergic and serotonergic circuits to regulate appetite, energy homeostasis, and anxiety. Clinically, HTR2C is a well-established factor in obesity and antipsychotic-induced weight gain. It is also implicated in mood and anxiety disorders and serves as a primary pharmacological target for several anti-obesity and psychotropic medications. |
